Leica TCS SP1 confocal workstation

The confocal scanhead is normally attached to an inverted Leica IMBE microscope and can also be used on an upright Leica RXA microscope. A custom-built cradle is available for sideways operation of the Leica RXA for gravity research. Both microscopes are equipped with prisms for differential interference contrast (DIC) and have a camera port with c-mount adapter.

  • 351 and 364 nm UV (Coherent, EnterpriseII 651)
  • 488 and 514nm Argon (Uniphase)
  • 561 nm Yellow diode-pumped solid state laser (Melles Griot 85-YCA)
  • 633 nm HeNe

The emission window of the 3 photomultipliers can be set to match the spectral properties of the fluorescent label. DIC images can be recorded simultaneously using the transmitted light detector and appropriate prisms. A free reader with basic data processing that runs on Windows XP can be obtained from the Leica ftp site (right click here and save installation file on your hard drive).

Leica RXA (upright)

  • support cradle for sideways operation (gravity research)
  • motorized z-control through RS port
